Abstract

EM maker claimed that EM is a solution of mixed culture of effective microorganisms capable of usefulness for the production of agricultural and forest products. It helps to control the balance of the environment I nature. It has also been used as a drink for some diseases remedy. It also consists of some Actinomycetes among the other microorganisms harboured in EM. Research on this group of bacteria was thus conducted with special attention to 2 important genera namely Actinomyces sp. and Streptomyces sp. In a pure culture isolation process used to determine for the Actinomyces sp., 3 samples of EM were done by using 2 selective media, but found no growth of this bacterial type. When these samples were tested for the Streptomyces sp. on Nutrient Agar. Sodium Caseinate Agar, Soil Extract Agar and Yeast-Extract Malt Extract Agar, a Streptomyces sp. was found to harbour with the sample designated as No.2 but confined only on Soil Extract Agar and Yeast Extract Malt Extract Agar, whose average total count was 8,502 CFU/ml. where as it was absent in sample No.1 and No.3. A pure culture isolation method was further conducted with this found genus which later revealed that it comprised of 6 isolates, designated as EM-STR 001, 002, 003, 004, 005 and 006. These isolated differed form one another in morphology and physiological activities when their characterizations were made by using the method of Bergey’s Manual of Bacteriology. The 6 isolates were further studied for their digesting abilities on filter paper, the results of which revealed that only 2 isolates i.e., EM-STR 001 and 005 could digest filter papers comparable with the digesting ability of Aspergillus niger (FPase = 0.034 U/ml) i.e., 0.030 U/ml respectively. Inhibition effects of the 6 isolates on the growth of Bacillus cereus, Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus and Asperggillus niger were also studied which found that only one isolate i.e., EM-STR 004 could inhibit the growth of Bacillus cereus as evidenced by inhibition zone of 1.5 cm. Other isolates could not inhibit any of the organisms tested.
